Would you prefer that we edit our old responses that are obsolete or otherwise deprecated with links to more recent relevant and accurate information that has been developed over time with more experience/work? If my aging memory is correct, I seem to recall seeing a blog a few years ago on here posted by the BR team saying the BR2 had been operated at 700 metres depth.

If that was so, what type of tether and what was used instead of the Fathom X which would have started to fail at 300 metres tether length?

I would also like to know if I stripped out a single twisted pair from the normal tether and and just used that alone, would I still get the benefit of being able to use it as the basis of a 300 metre tether instead of the 200 metres that the slim tether allows?


Metastatic castration-resistant prostate cancer (mCRPC) features high intratumoral cholesterol levels, due to aberrant regulation of cholesterol homeostasis. However, the underlying mechanisms are still poorly understood. The retinoid acid receptor-related orphan receptor gamma (RORγ), an attractive therapeutic target for cancer and autoimmune diseases, is strongly implicated in prostate cancer progression. We demonstrate in this study that in mCRPC cells and tumors, RORγ plays a crucial role in deregulation of cholesterol homeostasis. First, we found that RORγ activates the expression of key cholesterol biosynthesis proteins, including HMGCS1, HMGCR, and SQLE. Interestingly, we also found that RORγ inhibition induces cholesterol efflux gene program including ABCA1, ABCG1 and ApoA1. Our further studies revealed that liver X receptors (LXRα and LXRβ), the master regulators of cholesterol efflux pathway, mediate the function of RORγ in repression of cholesterol efflux. Finally, we demonstrated that RORγ antagonist in combination with statins has synergistic effect in killing mCRPC cells through blocking statin-induced feedback induction of cholesterol biosynthesis program and that the combination treatment also elicits stronger anti-tumor effects than either alone. Altogether, our work revealed that in mCRPC, RORγ contributes to aberrant cholesterol homeostasis by induction of cholesterol biosynthesis program and suppression of cholesterol efflux genes. Our findings support a therapeutic strategy of targeting RORγ alone or in combination with statin for effective treatment of mCRPC.

The present study has some limitations. First, this study was a real-world study, neither the chemotherapy regimen nor the ICI was consistent among patients. In addition, some patients received ICI alone while others received ICI plus chemotherapy. Due to the limited sample size, reliable comparisons between the two groups were difficult. Second, the status of PD-L1 expression on tumor cells or immune cells was unavailable at the time of this study, therefore, the impact of PD-L1 expression on the efficacy of immunotherapy was challenging to evaluate. Third, our study included previously heavily treated patients with a maximum of five lines of salvage chemotherapy, whether chemotherapy plus ICI as the first-line therapy would lead to a better outcome warrants further exploration. At present, we are conducting a prospective study to investigate the feasibility of postponing or omitting re-irradiation (RE) after tislelizumab (TI) plus chemotherapy (C) for unresectable (U), recurrent locoregionally advanced (LA) NPC (RETICULA-NPC, "type":"clinical-trial","attrs":"text":"NCT 04921995","term_id":"NCT04921995"NCT 04921995).


This can be a stressful, even scary, situation for college students. There are so many pressures from society that say eating alone is something to be ashamed of. In reality, eating alone is completely normal. According to a report in 2014 from The NPD Group, over 50% of eating happens when someone is alone.


Now there are 200,000 people who have left the city alone, altogether an estimated 400,000 at least have left other cities to head to the mountains and the countryside, and hundreds of them are living with almost no protection.


Time spent alone and feeling lonely can lead to depression. In fact, studies now show that a lonely brain is structurally and biochemically different. When someone is lonely, their neural responses to positive images and events get suppressed, so the world is perceived through a negative filter. We are more likely to believe that things are hopeless when we are lonely. This makes it more difficult to summon up the energy and bravery to find happiness and change.


While human beings need time alone to allow their brains to rest and rejuvenate, too much time alone or a lack of social connections can be harmful to our mental and physical health. It is important to distinguish between healthy time alone, where we are being productive, creative and introspective, versus negative time alone, where we are being self-critical or feeling lonely.


If you find yourself constantly around others and feeling depleted, make sure to schedule some healthy alone time. If you find yourself predominantly alone or are feeling lonely, make sure to invest more time seeking meaningful social connections. Whether you are an introvert or an extrovert, it is important to find the right balance for you.


After my aunt passed away, my cousin has been willing to move out alone. I understand that being alone helps to balance your brain, just like you mentioned it. He sure has a reason, so to show him my support, I will help him find a moving company to get things started.

This was a very interesting and helpful article and I really enjoyed the comments people shared about their lives and preferences. I do question if the people who are truly content with being alone are that way due to genetics, birth order, messages received and observed in the family of origin and expectations or a lack of expectations and the freedom to make their own choices. I also question what role these factors play in people who are more inclined to be introverts, both early in life and on, or after experiencing some losses and trauma or unspoken messages that they are not worthy unless they have someone in their life.


Division of labor is central to the ecological success of social insects. Among foragers of the honey bee, specialization for collecting nectar or pollen correlates with their sensitivity to sucrose. So far, differences in gustatory perception have been mostly studied in bees returning to the hive, but not during foraging. Here, we showed that the phase of the foraging visit (i.e. beginning or end) interacts with foraging specialization (i.e. predisposition to collect pollen or nectar) to modulate sucrose and pollen sensitivity in foragers. In concordance with previous studies, pollen foragers presented higher sucrose responsiveness than nectar foragers at the end of the foraging visit. On the contrary, pollen foragers were less responsive than nectar foragers at the beginning of the visit. Consistently, free-flying foragers accepted less concentrated sucrose solution during pollen gathering than immediately after entering the hive. Pollen perception also changes throughout foraging, as pollen foragers captured at the beginning of the visit learned and retained memories better when they were conditioned with pollen + sucrose as reward than when we used sucrose alone. Altogether, our results support the idea that changes in foragers' perception throughout the foraging visit contributes to task specialization.


Venezuelan equine encephalitis virus replicon particles (VRP) are infectious, non-propagating particles which function as a safe and robust humoral, mucosal and cellular adjuvant when codelivered with antigen. VRP are known to infect dendritic cells (DCs) in vitro and in vivo, and we have explored the role of DCs in the adjuvant activity of VRP. Upon footpad injection VRP are rapidly transported to the draining lymph node, resulting in cytokine secretion and recruitment of leukocytes. We found that DCs were preferentially infected in the lymph node, although other cell types were also infected. TNF-secreting monocyte-derived inflammatory DCs were the cell type most dramatically recruited to the lymph node and also the most readily infected by VRP. These inflammatory DCs also efficiently took up antigen. By depletion of DCs we demonstrated that both VRP transport to the lymph node and the subsequent inflammatory response were at least partly DC-dependent. By in vitro analysis we found that VRP-infected DCs can amplify CD4 and CD8 T cell responses to antigen, which is mediated at least partly by DC-secreted factors. We also show that VRP-infected DCs are sufficient generate an adjuvant effect comparable to that achieved by injection of VRP alone. Altogether, these data provide strong evidence that in vivo targeting of DCs by VRP matures and alters those DCs in a manner that potentiates the adjuvant activity of VRP.
